Short summary (English)

As part of the project Neurotransmitters and new mechanisms of drugs and toxins action dr. sc. Lidija Bach-Rojecky investigates pathophysiologic mechanisms of various types of chronic pain in experimental models of inflammatory, neuropathic and "mirror" pain. The current focus is investigation of the mechanisms of antinociceptive action of botulinum toxin type A and the possibility of its use in new indications. Various molecules involved in nociception (neurotransmitters, neuropeptides and their receptors) as well as markers of neuronal, astrocyte and microglia activation are determined in animal tissues (peripheral tissues, spinal cord, brain regions) using relevant biochemical methods (immunohistochemistry, Western blot, HPLC). The main focus of Petra Turčić, PhD research is in understanding the organoprotective and cytoprotective effect of short peptide sequences, with special emphasis on the application of the pro-opiomelanocortins fragments in inflammatory diseases of the digestive system. Drug research: Examination of melanocortin-alpha, melanocortin-beta, melanocortin-gamma, and other pro-opiomelanocortin peptide fragments in animal models of digestive system diseases. Studying the effect of melanocortin peptide by using their specific antagonists (eg HS024 and SHU9119). Modulation of the effect of melanocortin peptides by using their antisense (complementary) peptides according to the molecular recognition theory. The research of enantiomeric effects on biological activity and binding of pro-opiomelanocortin peptides, and the use of the molecular recognition theory in the modulation of their cytoprotective effects. The research group is currently investigating the ligand - receptor interaction of short bioactive peptide sequences using fluorescence spectroscopy.

U sklopu projekta Neurotransmitori i novi mehanizmi djelovanja lijekova i otrova dr. sc. Lidija Bach-Rojecky istražuje patofiziološke mehanizme nastanka različitih vrsta boli kroničnog trajanja, poput inflamatornih, neuropatskih boli, te refleksne boli na eksperimentalnim modelima. U užem znanstvenom interesu je istraživanje djelovanja botulinum toksina tipa A i mogućnosti njegove primjene u novim indikacijama. U tu se svrhu koriste relevantne biokemijske metode određivanja različitih neuronalnih (c-fos, p-CREb, pp38) i drugih staničnih markera uključenih u procese nocicepcije (određivanja neurotransmitora i neuropeptida, njihovih receptora) na animalnim tkivima (periferna tkiva, kralješnična moždina, regije mozga). Primarni istraživački interes dr.sc. Petre Turčić je proučavanje organoprotektivnog i citoprotektivnog učinka kratkih peptidnih sekvenci, s posebnim naglaskom na primjenu fragmenata pro-opiomelanokortina u upalnim bolestima probavnog sustava. Istraživanje lijekova: Ispitivanje melanokortina–alpha, melanokortina-beta, melanokortina-gama, te drugih peptidnih fragmenata pro-opiomelanokortina na animalnim modelima bolesti probavnog sustava.Ispitivanje učinka melanokortinskih peptida primjenom njihovih specifičnih antagonista (npr. HS024 i SHU9119). Modulacija učinka melanokortinskih peptida pomoću teorije molekularnog prepoznavanja korištenjem njihovih antisense (komplementarnih) peptida. Istraživanje učinaka enantiomerizacije na biološku aktivnosti i vezanje pro-opiomelanokotrinskih peptida, te primjena teorije molekularnog prepoznavanja u modulaciji njihovih citoprotektivnih učinaka. Istraživačka grupa trenutno istražuje ligand – receptor interakcije kratkih bioaktivnih peptidnih sekvenci pomoću fluorescenskijske spektroskopije.
