Short summary (English)

In Laboratory for Cell Technology, Application and Biotransformations research is focused on three scientific fields: animal cell technology, plants' biologically active compounds and ionic liquids as green solvents. Main focus in animal cell technology are bioprocesses for production of valuable biopharmaceuticals for therapeutic use. We were involved in the production of vaccines for veterinary use. To achieve better productivity of bioprocesses we optimized cell culture media and improved parameters, which influence cell growth and metabolism to obtaine higher biomass yields and cell viability of commercial cell lines (BHK, CHO). Also, optimization of different cultivating systems (T-flasks, spinner flasks, Wave bioreactor) was performed. Current research is in the field of environmental toxicology due to increased interest in analysis of toxic effects of numerous pharmaceutical and chemical agents accumulated in the aquatic environment. Our work includes development of model systems (fish cell lines) and methodologies for in vitro toxicology screening in order to minimize the use of in vivo acute fish bioassays. Our research interest is plants' secondary metabolites, glucosinolates and polyphenols, recognized for their health promoting properties. Main studies include the influence of cultivar and agro ecological cultivation conditions, and impact of some biotic and abiotic stresses on their composition and content. Also, we are evaluating plant extracts as ecologically acceptable and harmless corrosion inhibitors. Currently, the object of our studies is potential usage of plants for heavy metals phytoremediation. In recent years the increasing attention is focused on usage of ionic liquids, so called green solvents as potential replacement for volatile organic solvents in various processes. Current research interest is the synthesis of novel imidazolium- and pyridinium-based ionic liquids and their use as solvents in biocatalytic reactions. The emphasis is given to the development of environmentally friendly methods for ionic liquids preparation (solvent-free synthesis, microwave- and ultrasound-assisted synthesis).

U Laboratoriju za tehnologiju i primjenu stanica i biotrasfromacije istraživanja obuhvaćaju tri znanstvena područja: primjenu kulture životinjskih stanica, biljne biološki aktivne sastojke i ionske tekućine kao zelena otapala. Glavni cilj istraživanja u području primjene kulture životinjskih stanica su procesi za proizvodnju biofarmaceutskih proizvoda za terapeutsku primjenu. Aktivno se istraživao razvoj proizvodnje cjepiva za veterinarsku primjenu. U cilju bolje produktivnosti bioprocesa optimirani su sastav medija i parametri koji utječu na rast i metabolizam te su postignuti veći prinosi biomase i bolja vijabilnost proizvodnih staničnih linija (BHK, CHO). Također je optimirana primjena različitih sustava za uzgoj životinjskih stanica (T-boce, spinner boce, Wave bioreaktor). Trenutna istraživanja usmjerena su ka području toksikologije okoliša zbog povećanog zanimanja za analize toksičnih učinaka brojnih farmaceutika i kemikalija koje se nakupljaju u vodenim sustavima. Stoga, istraživanja obuhvaćaju razvoj modelnih sustava (stanične linije riba) i metodologija za primjenu kultura stanica u in vitro toksikološkim analizama u cilju smanjenja in vivo pokusa na ribama. Biljni biološki aktivni sastojci (fitokemikalije) na samom su vrhu svjetskih istraživanja kojima je cilj poboljšanje uzgoja biljnih kultura s definiranim i povećanim udjelom kemozaštitnih spojeva. Istraživanja obuhvaćaju utjecaj sorte i agroekoloških uvjeta te učinak nekih biotičkih i abiotičkih stresova na sastav i udio biološki aktivnih metabolita (polifenoli, glukozinolati). Također se provodi vrednovanje biljnih ekstrakata kao ekološki neškodljivih inhibitora korozije. Trenutni interes grupe je usmjeren na ispitivanja mogućnosti uporabe biljaka za fitoremedijaciju teških metala iz tla. Posljednjih je godina sve veća pozornost znanstvenika usmjerena ka uporabi ionskih tekućina, tzv. „zelenih“ otapala, kao potencijalnoj zamjeni za hlapljiva organska otapala u raznim procesima. Aktivnost grupe uključuje pripravu novih imidazolijevih i piridinijevih ionskih tekućina te njihovu primjenu kao otapala u biokatalitičkim reakcijama. Naglasak istraživanja je na razvoju ekološki prihvatljivih metoda priprave ionskih tekućina (sinteza bez prisustva štetnih otapala te sinteza potpomognuta mikrovalnim i ultrazvučnim zračenjem).
